Abstract: Resumen .- En la presente investigación se obtuvieron extractos clarificados y concentrados con actividad inhibidora frente a papaína y quimotripsina, provenientes de las semillas de amaranto, arveja, chocho, fréjol y sangorache. Se prepararon extractos crudos con las harinas de las semillas desengrasadas. Se determinó la actividad inhibidora específica (AIE) de los extractos crudos frente a papaína, quimotripsina y carboxipeptidasa A. Los mejores resultados se obtuvieron con fréjol frente a papaína (4,95 U/mg) y con chocho frente a quimotripsina (2,19 U/mg). Los extractos seleccionados fueron tratados con ultrafiltración centrífuga a 10 kDa y tratamiento calórico lo que produjo un grado de purificación de 2,91 y 3,02 veces en cada extracto. Se determinaron los pesos moleculares de los inhibidores mediante electroforesis SDS-PAGE, cuyos resultados fueron 5,18 y 4,05 kDa para los inhibidores de chocho y fréjol, respectivamente. Los valores de IC50 fueron 0,018 y 0,107 mM, frente a quimotripsina y papaína. Se estudió la estabilidad a tres temperaturas -20, 4 y 17 °C, durante cuatro semanas. Los mejores resultados se obtuvieron a -20 °C. Se diseñó una planta piloto para la obtención de 5 L/d de extracto inhibidor y se determinó el costo de producción en $ 0,134/mg. Abstract .- In the present investigation extracts with inhibitory activity against papain and chymotrypsin, were clarified and concentrated from amaranth, peas, lupine, beans and sangorache seeds. Crude extracts were prepared with the defatted seed meal. Specific inhibitory activity (SIA) of crude extracts against papain, chymotrypsin and carboxypeptidase A. The best results were obtained with beans against papain (4.95 U / mg) and lupine against chymotrypsin (2.19 U / mg). Selected extracts were treated with 10 kDa centrifugal ultrafiltration and heat treatment which produced a degree of purification of 2.91 and 3.02 times in each extract. The molecular weights of inhibitors by SDS-PAGE electrophoresis and the results were 5.18 and 4.05 kDa for inhibitors of lupine and beans, respectively. The IC50 values were 0.018 and 0.107 mM against chymotrypsin and papain, respectively. Stability at three temperatures -20, 4 and 17 ° C was studied for four weeks. The best results were obtained at -20 ° C. A pilot plant for the production of 5 L / d of inhibitor extract was designed and the cost of production was determined at $ 0.134 / mg.
