Zusammenfassung: In this study, the methodology developed for the quantification of nematic cell polarity in three-dimensional tissues was applied to the case of human liver tissue under two relevant physiological conditions: healthy tissue, and tissue with Non-Alcoholic Fatty Liver (NAFLD) pathology. The study shows that human liver tissue behaves like a biaxial liquid crystal with localized defects. For the case of human samples with NAFLD, a decrease in repulsion between the apical and basal surfaces was observed compared to the case of healthy tissue. To better understand the mechanisms underlying the liquid crystalline order in human liver tissue, several coordination models of biaxial nematic polarity were analyzed using Monte Carlo simulations. The simulations showed that in the coordination of the hepatocytes polarity with an external field (probably generated by the surrounding extracellular matrix), the local cell-cell interactions and the existence of point defects are necessary to generate configurations like those observed in the tissue.
Beschreibung: En este estudio, se aplicó la metodología desarrollada para la cuantificación de polaridad celular nemática en tejidos tridimensionales, al caso del tejido hepático humano bajo dos condiciones fisiológicas relevantes: tejido sano, y tejido con la patología de Hígado Graso No Alcohólico (HGNA). El estudio muestra que el tejido hepático humano se comporta como un cristal líquido biaxial con defectos localizados. Para el caso de muestras humanas con HGNA, se observó una disminución en la repulsión entre las superficies apical y basal respecto al caso de tejido sano. Para comprender mejor los mecanismos subyacentes al orden líquido cristalino en tejido hepático humano, se analizaron varios modelos de coordinación de la polaridad nemática biaxial mediante simulaciones Monte Carlo. Las simulaciones mostraron que en la coordinación de la polaridad de los hepatocitos con un campo externo (probablemente generado por la matriz extracelular circundante), las interacciones locales célula-célula y la existencia de defectos puntuales son necesarias para generar configuraciones como las observadas en el tejido.
