Abstract: The main purpose of this research was to obtain peptidic hydrolysates with antioxidant activity from gelatin extracted from rabón bueno skin (Alopias pelagicus). The experiment was carried out in two phases. In the first one, a completely randomized design was used having the extraction temperature as the variable in levels of 55, 60, 65 and 70 °C. Obtaining the best results at 65 °C, based on the analysis of yield (41,41%) gel strength (0,016 ± 0.002 kN) melting and gelling point (28 ° C and 22 ° C), apparent viscosity (70,519 Pa.s) and protein (88,34%). From this extraction the second phase begun with a completely randomized design with the hydrolysis times of 15, 30, 45, 60, 120 and 180 minutes with the proteolytic enzyme GRANOZYME.ACC. Based on the analysis of antioxidant activity by ABTS, it was determined that the hydrolysis at 180 minutes, gave peptides with higher activity (0,33 mg ascorbic acid Eq/g.protein). This sample was fractionated into 3, 10, 50 and 100 kDa. In each fraction the antioxidant activity was evaluated by the methods: ABTS, FRAP and DPPH. The best result of ATBS was reached with the 3 kDa fraction (0,12 M ascorbic acid Eq/g.protein). Whereas, in DPPH analysis in that of 100 kDa (1,98 mM ascorbic acid Eq/g.protein) and by the FRAP method the best result was obtained from the hydrolyzed gelatin (16,46 μM.Eq.FeSO4/g.protein).
Description: La presente investigación tuvo como objetivo la obtención de hidrolizados peptídicos con actividad antioxidante a partir de gelatina extraída de piel de rabón bueno (Alopias pelagicus). El experimento se realizó en dos fases, en la primera, se empleó un diseño completamente al azar teniendo como variable la temperatura de extracción con niveles de 55, 60, 65 y 70 °C, obteniéndose los mejores resultados a 65 °C, eso tras los análisis de rendimiento (41,41 %) fuerza del gel (0,016 ± 0,002 kN), temperatura de fusión y gelificación (28 °C y 22 °C), viscosidad aparente (70,519 Pa s), y proteína (88,34 %). A partir de esta extracción se procedió a la segunda fase en la cual se llevó a cabo un diseño completamente al azar con tiempos de hidrólisis de 15, 30, 45, 60, 120 y 180 min con la enzima proteolítica GRANOZYME.ACC. Tras el análisis de actividad antioxidante por ABTS, se determinó que la hidrólisis a 180 minutos originó péptidos con mayor actividad (0,33 mg de ac. ascórbico Eq/g.de.proteína). Esta muestra se fraccionó en 3, 10, 50 y 100 kDa. En cada fracción se evaluó la actividad antioxidante por los métodos: ABTS, FRAP y DPPH. El mejor resultado de ATBS se alcanzó con la fracción de 3 kDa (0,12 M ac. ascórbico Eq/g.de.proteína), mientras que en el análisis por DPPH en aquella de 100 kDa (1,98 mM ac. ascórbico Eq/g.de.proteína) y por el método FRAP el mejor resultado se obtuvo de la gelatina hidrolizada (16,46 μM.Eq.FeSO4/g.de.proteína).
