dc.descriptionEl biorreactor tipo tanque agitado LiFlus GX ubicado en el Laboratorio de Bioprocesos posee un sistema de acondicionamiento de temperatura ineficiente, lo que evita que durante la fase de adaptación un microorganismo alcance su temperatura óptima de crecimiento. En este trabajo se evalúa el desempeño del sistema de acondicionamiento de temperatura en el biorreactor mediante modelado matemático con la incorporación de agua de calentamiento y un aro de neopreno como aislante en la chaqueta. Se midieron las dimensiones del equipo para establecer parámetros de operación como área de transferencia de calor, volumen de trabajo y potencia de agitación. Se analizó el comportamiento de la niquelina como actuador del sistema de acondicionamiento para establecer su capacidad operativa. El modelo matemático se formuló en el contexto de un proceso fermentativo aerobio y se validó mediante pruebas experimentales con Saccharomyces cerevisiae marca DistilaMax® en un medio de YPD al 5 % de glucosa. Se determinó que sin fluido de calentamiento el espacio entre la chaqueta y el recipiente de vidrio es un reservorio de aire que actúa como aislante térmico y expone a la resistencia a sobrecalentamiento. El agua de calentamiento permite el desarrollo de fermentaciones a temperatura óptima con duración de 450 min sin interrupciones y con la niquelina operativa con 0,21 % de su capacidad. La fase de adaptación de la levadura Saccharomyces cerevisiae, donde el efecto de la temperatura es marcado, se describe con mayor aproximación mediante el modelo de Hills y Wright, con una desviación de apenas 4,25 % cuando no se utilizó empaque de neopreno y del 4,60 % cuando sí se lo utilizó. Además, el uso del empaque de neopreno minimiza las pérdidas de calor, y en consecuencia reduce el tiempo de la fase de latencia en 30 min.es_ES
dc.description.abstractThe LiFlus GX stirred tank bioreactor located in the Bioprocess Laboratory has an inefficient temperature conditioning system, which prevents a microorganism from reaching its optimum growth temperature during the adaptation phase. In this work, the performance of the temperature conditioning system in the bioreactor is evaluated through mathematical modeling with the incorporation of heating water and a neoprene ring as insulation in the jacket. The dimensions of the equipment were measured to establish operating parameters such as heat transfer area, work volume and stirring power. The behavior of nickeline as an actuator of the conditioning system was analyzed to establish its operational capacity. The mathematical model was formulated in the context of an aerobic fermentative process and validated through experimental tests with Saccharomyces cerevisiae brand DistilaMax® in a YPD medium at 5% glucose. It was determined that without heating fluid, the space between the jacket and the glass container is an air reservoir that acts as a thermal insulator and exposes the resistance to overheating. The heating water allows the development of fermentations at optimum temperature with a duration of 450 min without interruptions and with operative nickeline with 0.21% of its capacity. The adaptation phase of the Saccharomyces cerevisiae yeast, where the effect of temperature is marked, is more closely described by the Hills and Wright model, with a deviation of only 4.25 % when neoprene packing was not used, and the 4.60% when it was used. In addition, the use of neoprene packing minimizes heat losses, and consequently reduces the lag phase time by 30 min.es_ES
